Overview

Fc gamma receptor IIb (FCGR2B) is a low affinity inhibitory receptor for the Fc region of immunoglobulin gamma (IgG), uniquely distinguished by the presence of an immunoreceptor tyrosine-based inhibitory motif (ITIM) in its cytoplasmic region. It is primarily expressed on B cells, monocytes, basophils, and dendritic cells, where it regulates immune responses by inhibiting cellular activation, promoting phagocytosis of immune complexes, and down-regulating antibody production. There are distinct isoforms generated through alternative splicing which differ in their cellular localization and regulatory functions. Genetic polymorphisms or reduced function of FcγRIIb are linked to susceptibility to various autoimmune diseases. This receptor is being investigated as a therapeutic target in autoimmunity and oncology, both to suppress harmful immune activation and as part of engineered antibody therapies.
