Overview

Low affinity immunoglobulin gamma Fc region receptor III-A (CD16a) is a transmembrane glycoprotein expressed on NK cells, macrophages, and some monocytes. It binds to the Fc portion of IgG antibodies clustered on target cells, triggering antibody-dependent cellular cytotoxicity (ADCC) and other immune responses. Genetic polymorphisms, particularly F158/V158, affect IgG binding affinity and clinical efficacy of therapeutic antibodies. After activation, CD16a surface expression is downregulated through proteolytic shedding.
