Target intelligence / Profile preview

Low affinity immunoglobulin gamma Fc region receptor III-B (CD16b)

Target
CD16b
Molecular classification
Receptor (specifically an immunoglobulin G [IgG] Fc receptor), Cell surface protein anchored via glycosylphosphatidylinositol (GPI) linkage
01

Overview

The Low affinity immunoglobulin gamma Fc region receptor III-B (CD16b, encoded by the FCGR3B gene) is a cell surface glycoprotein expressed primarily on human polymorphonuclear neutrophils. It binds the constant region (Fc) portion of immunoglobulin G (IgG), exhibiting low affinity compared to other family members. Unlike its close homolog CD16a/FcγRIIIA—which mediates antibody-dependent cellular cytotoxicity—CD16b does not trigger phagocytosis or cytotoxic responses upon binding antibodies. Instead, it acts mainly as a "trap" for circulating immune complexes without activating downstream inflammatory signaling pathways in neutrophils. This unique function helps regulate peripheral immunity and prevent excessive tissue damage from inappropriate activation during normal clearance processes. Variations or deficiencies in FCGR3B have been linked to altered susceptibility to infection and autoimmunity; thus it represents both a biomarker candidate and potential therapeutic target within the broader context of inflammatory disease management.

Mechanism of action

For drugs targeting related pathways or using engineered antibodies with altered effector functions: Modulation of immune complex clearance. Potential reduction of inflammation by preventing inappropriate activation of neutrophils. Alteration of antibody effector functions through engineering to avoid binding to this low-affinity receptor.

Biological functions

Immune response modulationBinding to complexed or aggregated IgG and monomeric IgGClearance/trapping of immune complexes in peripheral circulation without activating neutrophils
04

Disease associations

Inflammation (notably in autoimmune and inflammatory diseases)Infection susceptibility/modulationNeonatal alloimmune neutropenia, paroxysmal nocturnal hemoglobinuria, other disorders involving neutrophil dysfunction or immune complex handling
05

Safety considerations

Therapeutic targeting could potentially impair clearance of circulating immune complexes, increasing risk for infections or autoimmunity if not carefully managed.Genetic deficiency is associated with increased susceptibility to certain infections but also reduced risk for some autoimmune complications.
06

Interacting drugs

There are currently no approved drugs that directly target CD16b/FCGR3B specifically. However, its close homologs such as CD16a/FcγRIIIA are targeted by some monoclonal antibody therapies for their role in antibody-dependent cellular cytotoxicity. Drugs modulating the broader class of IgG receptors may have indirect effects.

Biomarkers

Expression levels can be used as biomarkers for certain autoimmune/inflammatory conditions.Used experimentally as a marker for specific subsets of neutrophils and granulocytes in flow cytometry panels.
