ΔLNGFR (truncated Low-Affinity Nerve Growth Factor Receptor), also known as truncated CD271 or p75NTR, is a modified version of the human nerve growth factor receptor that lacks the intracellular signaling domain [1]. It is extensively used in adoptive immunotherapy and gene therapy as a non-immunogenic surface marker to identify, enrich, and track transduced cells, such as CAR-T cells or hematopoietic stem cells [2]. Because the cytoplasmic signaling tail is removed, the molecule cannot transmit biological signals, preventing it from interfering with the host cell's function or promoting unintended cellular responses [3]. In clinical applications, ΔLNGFR allows for the high-purity selection of engineered cells using clinical-grade magnetic-activated cell sorting (MACS) [4]. Additionally, it can serve as a target for antibody-mediated depletion of engineered cells in the event of severe adverse effects, acting as a component of a safety switch system [5].
