Low-density lipoprotein oxidation is the process by which native circulating LDL particles undergo chemical modifications—primarily through lipid peroxidation and protein alteration—rendering them proatherogenic. This modified form contributes directly to the initiation and progression of atherosclerotic lesions by promoting foam cell formation, inflammation, endothelial dysfunction, plaque instability, and ultimately increasing cardiovascular disease risk. Both extracellular mechanisms involving reactive intermediates from vascular cells as well as intracellular pathways mediated by iron-catalyzed reactions inside macrophage lysosomes are implicated in this critical pathological event.
