Overview

Low-density lipoprotein receptor class A domain-containing protein 3 (LDLRAD3) is a conserved cell-surface protein that belongs to the LDL receptor family and contains a characteristic LDL receptor type-A module. It is expressed in neurons, epithelial cells, myeloid cells, and muscle. LDLRAD3 acts as a recently identified receptor for Venezuelan equine encephalitis virus (VEEV), mediating viral attachment and entry in host cells. Biochemical studies indicate it is involved in receptor-mediated endocytosis and may regulate amyloid precursor protein (APP) processing, potentially influencing amyloid-beta peptide production, which has roles in neurodegenerative pathology. The protein is further predicted to regulate E3 ubiquitin ligase activity and may modulate protein degradation pathways. The endogenous ligand(s) for LDLRAD3 remain unknown, and no approved drugs are currently known to target this protein directly.
