Overview

Low-density lipoprotein receptor-related protein 11 (LRP11) is a membrane-bound receptor in the LDL receptor family, expressed in multiple tissues. LRP11 is highly expressed in certain cancer cells, notably hepatocellular carcinoma, where it promotes cell proliferation, migration, and invasion. Mechanistically, LRP11 acts as a scaffold protein, facilitating the interaction and deubiquitination of RACK1 (a signaling scaffold) by USP5, thereby stabilizing RACK1 and supporting malignant progression. LRP11 also interacts with the tumor microenvironment, showing positive correlations to infiltration by diverse immune cells (such as macrophages, neutrophils, T and B lymphocytes). Hypomethylation of the LRP11 promoter is a major route for its upregulation in tumors and is considered a promising prognostic biomarker. Overall, LRP11 is being actively researched as a molecular target and prognostic indicator in liver cancer and possibly other malignancies.\n\nKey findings include its function in supporting tumor growth and immune infiltration, making it a candidate for future targeted therapeutics and diagnostics. However, clinical translation, including safety and efficacy data for LRP11-directed agents, has not yet been established.
