Overview

The loxP site is a specific 34-base-pair DNA sequence derived from the P1 bacteriophage and is the recognition and recombination site for Cre recombinase. It consists of two 13 bp inverted repeats that flank an 8 bp spacer region, which gives the site its directionality. Cre recombinase catalyzes site-specific recombination at loxP sites, enabling precise deletions, insertions, or inversions of DNA between two loxP sites. This forms the basis of the widely used Cre-lox system in genetic engineering, especially for generating tissue-specific or conditional gene modifications in model organisms[1][3][4][6].
