Overview

LRRN4 C-terminal-like protein (LRRN4CL) is a single-pass membrane protein and a member of the leucine-rich repeat neuronal (LRRN) family, encoded by the LRRN4CL gene (UniProt Q8ND94)[2][3]. LRRN4CL is predicted to localize to the membrane and is structurally related to proteins with roles in cellular adhesion and signal transduction. It is overexpressed in colorectal cancer (CRC) and acts as an independent prognostic factor for disease progression and survival. Experimental data show that LRRN4CL promotes cancer cell proliferation, enhances DNA synthesis, accelerates cell migration, and suppresses apoptosis, likely through modulation of the Ras/MAPK and PI3K signaling pathways[1]. There is also a possible link to Charcot-Marie-Tooth disease, though the pathophysiological significance remains to be clarified[3]. No drugs are currently described as interacting with LRRN4CL, and it is not classified within traditional drug target families (e.g., GPCR, enzyme, transporter), but its expression is a candidate biomarker for poor prognosis in CRC[1][3].
