LTO1 maturation factor of ABCE1 (often simply called LTO1) is a protein involved in the maturation of the highly conserved eukaryotic protein ABCE1 (also called Rli1 in yeast) by mediating the insertion of [4Fe-4S] clusters into ABCE1[Rli1][1][2][3]. LTO1 forms a dedicated adaptor complex with YAE1D1 (Yae1) to specifically recruit ABCE1 to the cytosolic iron-sulfur (Fe-S) cluster assembly (CIA) machinery. This process is essential for ribosome biogenesis, translational initiation, termination, and recycling. Unlike more generalized maturation factors, LTO1 function is highly specific and operates downstream of core CIA components. Its human orthologs are ORAOV1 and TAOS1, and LTO1 can be overexpressed in certain cancers such as oral squamous cell carcinoma[3][1]. There are currently no known pharmacological agents or drugs directly targeting LTO1, nor accepted diagnostic or predictive biomarkers based on LTO1 status, and its role as a therapeutic target remains unestablished[3].
