Overview

The LUC7-like family comprises three human proteins (LUC7L, LUC7L2, LUC7L3) that are conserved nuclear zinc finger-containing splicing factors. They are fundamental components of the spliceosome and function primarily to regulate pre-mRNA splicing by interacting with small nuclear RNAs (notably U1 snRNA) and specifying 5' splice-site selection. Each member has a somewhat distinct binding pattern on pre-mRNAs and tissue-specific expression, enabling fine-tuned regulation of constitutive and alternative splicing. LUC7-like proteins are essential for normal cell function, differentiation, and play a critical role in hematopoietic cells. Dysfunction or loss, especially of LUC7L2, is linked to impaired splicing and certain leukemias.

Mechanism of action

Modulation of pre-mRNA splicing, mainly by influencing recognition and usage of 5' splice sites when targeted by splicing regulatory molecules

03

Biological functions

Regulation of alternative splicingmRNA processingAssociation with spliceosomal complexesControl of 5' splice-site selection
04

Disease associations

Cancer (especially acute myeloid leukemia via LUC7L2 haploinsufficiency/mutations)Hematopoietic differentiationOther diseases potentially involving splicing dysregulation
05

Safety considerations

Currently none described as a direct drug targetModulation of splicing factors may disrupt normal gene expression and be toxic to non-cancerous cells
06

Biomarkers

LUC7L2 loss or mutation could serve as a biomarker in some myeloid malignancies
