Overview

Luminal and mucosal proteins encompass a broad spectrum of proteins located within the lumen and the mucosal layer of the gastrointestinal tract and other mucosal surfaces. This group includes digestive enzymes, transport proteins, mucins, and plasma proteins that may be present due to exudation at sites of injury (StatPearls, 2023). These proteins serve as the functional targets for mucosal protectants and cytoprotective agents, which are used to treat conditions like peptic ulcer disease and gastroesophageal reflux disease. For instance, the drug sucralfate binds to positively charged proteins such as albumin and fibrinogen at the site of an ulcer, forming a protective physical barrier against aggressive factors like gastric acid and pepsin (DrugBank, 2024). Beyond their role in drug binding, these proteins are essential for nutrient absorption, immune defense, and maintaining the integrity of the mucosal barrier (World J Gastroenterol, 2007). Dysfunction or degradation of these proteins is often implicated in inflammatory and ulcerative diseases of the gut. Consequently, therapeutic strategies often focus on protecting these proteins or utilizing them to deliver localized treatments.
