The luminal contents in the gastrointestinal tract refer to the heterogeneous mixture of ingested food, water, digestive secretions, and microorganisms located within the hollow interior of the digestive organs (NIDDK, 2023). While not a specific molecular target like a protein or receptor, the luminal environment is a significant site for pharmacological action, particularly for drugs that do not require systemic absorption to be effective (StatPearls, 2023). Similarly, activated charcoal is utilized in emergency medicine to adsorb toxins and drugs within the stomach and intestines, thereby reducing systemic toxicity (StatPearls, 2023). The composition of the luminal contents is also critical for maintaining the gut microbiota, which influences overall metabolic and immune health (NIH, 2012). Therapeutic strategies targeting this compartment often focus on physical sequestration, osmotic modulation, or the inhibition of enzymes like lipases that function within the lumen (PubMed, 2000).
Drugs targeting the luminal contents typically act through physical adsorption, chemical sequestration or chelation, osmotic retention of water, or by inhibiting enzymes that act on luminal substrates (StatPearls, 2023; PubMed, 2000).
