Overview

LY6E divergent transcript (LY6E-DT, also known as LOC100133669) is a human gene originally annotated as a long non-coding RNA. Recent research shows it encodes a conserved 153-amino-acid microprotein called Metastatic-Related Protein (MRP). LY6E-DT and MRP are highly expressed in metastatic breast cancer tissues and cell lines. Functionally, LY6E-DT regulates breast cancer invasion and metastasis by two main mechanisms: the microprotein MRP binds to HNRNPC, increasing the stability and expression of EGFR mRNA, which activates PI3K/AKT signaling, and the lncRNA itself promotes nuclear import of the transcription factor YBX1, leading to activation of ZEB1—a master regulator of EMT and cell migration. MRP expression also shows potential as a biomarker for lymph node metastasis in breast cancer. There are no known drugs targeting LY6E-DT or MRP, and it is not considered a classical therapeutic target such as a receptor, enzyme, or transporter as of current knowledge[2]. Note: LY6E-DT is a distinct gene from LY6E. LY6E encodes a GPI-anchored surface protein with antiviral and immune-related functions, and is a different molecular entity[1]. LY6E-DT is not a receptor, enzyme, or transporter, but a gene encoding an lncRNA and a microprotein involved in cancer biology.
