Overview

The lymph node reticular meshwork is a specialized structural and functional scaffold within the lymph node, primarily composed of fibroblastic reticular cells (FRCs) and an extracellular matrix (ECM) rich in collagen type III (Fletcher et al., 2015, Nature Reviews Immunology). It provides a physical track for the rapid migration of T cells and dendritic cells, facilitating the efficient scanning of antigens (Bajénoff et al., 2006, Immunity). Additionally, the meshwork forms a conduit system that allows for the rapid transport of low-molecular-weight lymph-borne molecules, such as chemokines and small antigens, directly to the paracortex (Roozendaal et al., 2009, Immunity). In pathological conditions like chronic infection or cancer, the meshwork can undergo extensive remodeling or fibrosis, which impairs immune cell trafficking and contributes to immune exhaustion or metastasis (Acton et al., 2014, Nature). While the meshwork itself is an anatomical structure rather than a single molecular target, its components—including FRC-derived chemokines like CCL19 and CCL21—are critical for orchestrating the adaptive immune response and are investigated as therapeutic focal points (Link et al., 2007, Nature Immunology).
