Lymphatic endothelial transcriptional regulator lncRNA 1 (LETR1) is a long non-coding RNA specifically and robustly expressed in lymphatic endothelial cells. LETR1 operates primarily within the cell nucleus as a trans-acting lncRNA, serving as a scaffold for epigenetic regulators that fine-tune the transcription of genes essential for lymphatic endothelial growth and migration, including *KLF4* and *SEMA3C*. Identified under the systematic names LINC01197 and CTD-2536I1.1, LETR1 belongs to the intergenic lncRNA family, functioning as a critical regulator of vascular identity and homeostasis. LETR1 does not encode a protein nor serve as a receptor, but its cell-type specificity makes it a potential biomarker for LECs and a plausible candidate in vascular developmental diseases.
