Overview

Lymphatic vessels and sentinel lymph node tissue constitute the anatomical network responsible for fluid homeostasis, lipid transport, and immune surveillance (NIH, 2023). In the context of oncology, the sentinel lymph node (SLN) is defined as the hypothetical first lymph node or group of nodes reaching which cancer cells are most likely to spread from a primary tumor (StatPearls, 2023). This tissue is not a single molecular target but rather a physiological system targeted by diagnostic tracers and imaging agents to facilitate sentinel lymph node biopsy (SLNB). Identifying the SLN is crucial for accurate cancer staging and determining the necessity of more extensive lymph node dissection in diseases like breast cancer and melanoma (NCI, 2022). Diagnostic agents such as Technetium Tc-99m tilmanocept specifically target receptors like CD206 within the lymph node to facilitate visualization (FDA, 2013). Therapeutic challenges associated with this system include the risk of lymphedema following surgical intervention and the potential for allergic reactions to mapping dyes (PubMed, 2021).
