Overview

Lymphatic vessels and sentinel lymph node tissues represent anatomical structures rather than a single molecular target. Lymphatic vessels are a network of thin-walled capillaries and vessels that maintain tissue fluid homeostasis and facilitate the transport of immune cells and antigens to lymph nodes (StatPearls, 2023). The sentinel lymph node (SLN) is defined as the first lymph node or group of nodes draining a primary tumor, serving as the initial site for metastatic seeding (National Cancer Institute, 2023). In clinical oncology, these tissues are the focus of diagnostic mapping procedures using agents like Technetium Tc-99m tilmanocept or Indocyanine green to guide surgical staging in cancers such as melanoma and breast cancer (PubMed, PMID: 25107425). While the tissues themselves are not therapeutic targets in the traditional sense, they express specific molecular markers like Lymphatic Vessel Endothelial Hyaluronan Receptor 1 (LYVE-1) and Vascular Endothelial Growth Factor Receptor 3 (VEGFR-3) that are investigated for their roles in lymphangiogenesis and tumor progression (UniProt, P35916).

Mechanism of action

Diagnostic agents are administered to map lymphatic drainage and identify the sentinel lymph node through receptor-mediated binding (e.g., tilmanocept binding to CD206) or passive flow and entrapment within the lymphatic network.
