Lymphocyte antigen 6 complex locus C (Ly6C) is a cell surface glycoprotein belonging to the Ly6/uPAR superfamily, anchored by a GPI-link, and primarily studied in mice. It is prominently expressed on subsets of immune cells, including monocytes, macrophages, neutrophils, dendritic cells, and select T-cell populations[5]. Ly6C is used extensively to distinguish between monocyte subpopulations in mice: Ly6C^high^ monocytes are considered pro-inflammatory and are rapidly recruited to sites of infection or injury, while Ly6C^low^ monocytes display patrolling and anti-inflammatory functions[5]. In experimental models, Ly6C^+^ monocytes play a pivotal role in immune defense, influencing the outcomes of infections (such as hepatitis B), inflammation, and tissue repair[6]. Ly6C expression is a key biomarker for immune cell subset identification and serves as a functional marker in studies of inflammation, infection, and immune-mediated diseases[5][6][4]. In cancer, human Ly6 family members do not have a direct Ly6C homolog but are considered biomarkers associated with poor prognosis and immune regulation in tumors[3]. Ly6C is not currently a direct drug target, but modulation of Ly6C^+^ populations is a potential avenue for immunotherapeutic strategies[6]. While "Lymphocyte antigen 6 complex, locus C" (Ly6C) is well-characterized in mouse immunology, there is no one-to-one human homolog, though analogous Ly6 superfamily genes exist in humans[3][5][7].
