Overview

Lymphocyte antigen 6 complex locus C1 (Ly6C) is a GPI-anchored cell-surface protein encoded by the Ly6c1 gene in mice. It belongs to the Ly6/uPAR superfamily, characterized by a conserved LU domain with multiple cysteine residues forming disulfide bridges and a “three-finger” protein topology similar to snake venom neurotoxins[5]. Ly6C is used extensively as a cell surface marker to distinguish subsets of immune cells, especially monocytes, T cells, and dendritic cells. High Ly6C expression (Ly6C^hi) marks pro-inflammatory monocytes, while low Ly6C (Ly6C^lo) defines patrolling monocytes. Ly6C is also expressed on subsets of memory CD8+ T cells and is upregulated during inflammation. Its main utility is in murine immunology and preclinical disease models for cell tracking and functional assays but it is not a druggable human therapeutic target[5][6][7].
