Lymphocyte antigen 6 complex locus G (Ly-6G) (Ly-6G)

Target
Ly-6G
Molecular classification
Cell-surface antigen, GPI-anchored glycoprotein, Member of Ly-6/uPAR protein family
01

Overview

Lymphocyte antigen 6 complex locus G (Ly-6G) is a GPI-linked glycoprotein, 21–25 kDa, predominantly expressed on mature mouse neutrophils, serving as an essential marker for their identification and functional state; it plays roles in neutrophil migration through interaction with β2 integrins, in myeloid maturation, and in immune responses to infection and inflammation. Ly-6G is used as a target for experimental depletion of neutrophils in vivo, and its expression correlates with neutrophil aging and functional impairment in animal models. In humans, the functional homolog is CD177 and not Ly-6G. Members of the Ly-6 family are involved in diverse processes including cell proliferation, migration, and immune defense.

02

Mechanism of action

Anti-Ly-6G antibody binds Ly-6G on neutrophils, potentially leading to their depletion via immune-mediated mechanisms or blockade of migration/function

03

Biological functions

Neutrophil differentiation and identificationImmune responseRegulation of neutrophil migration (via interaction with β2 integrins)Myeloid cell maturationMarker for neutrophil function in aging, inflammation, and infection
04

Disease associations

Inflammation (role in migration and recruitment of neutrophils during inflammatory responses)Infection (marker and mediator of neutrophil responses to bacterial and parasitic infections)Cancer (Ly-6 family members implicated in tumorogenesis)Immune aging (marker for neutrophil functional decline in aging)
05

Safety considerations

In mouse models, anti-Ly-6G treatments can cause profound neutropenia, elevating risk of infection and impairing innate immunityNot directly applicable to humans due to lack of true Ly-6G homolog
06

Interacting drugs

anti-Ly-6G antibody (clone 1A8)
07

Biomarkers

Ly-6G expression level is used as a biomarker for neutrophil population quantification and functional statusmonitoring age-related decline in neutrophil function in mice
