Overview

Lymphocyte antigen 6 complex locus G (Ly6G) is a 21-25 kDa glycosylphosphatidylinositol (GPI)-anchored cell surface protein specifically expressed on mature murine neutrophils (UniProt P35461). As a member of the Ly6/uPAR superfamily, it serves as a definitive marker for identifying and isolating neutrophils in mouse models (PubMed: 23365456). Functionally, Ly6G is involved in modulating neutrophil migration and recruitment to inflammatory sites, likely through its association with the beta-2 integrin CD11b/CD18 (PubMed: 22585682). In preclinical research, Ly6G is a primary target for neutrophil depletion using monoclonal antibodies like clone 1A8, which allows researchers to study the specific contribution of neutrophils to diseases such as sepsis, cancer, and autoimmune disorders (PubMed: 21930770). While Ly6G is a crucial tool in murine immunology, it lacks a direct human ortholog, which presents a challenge for direct clinical translation but remains vital for understanding innate immune mechanisms (PubMed: 24610276). Targeting this molecule effectively modulates the innate immune response by reducing the number of circulating and infiltrating neutrophils.
