Overview

Lymphocyte antigen 6 complex locus protein G6f (LY6G6F) is a type I transmembrane protein that belongs to the immunoglobulin superfamily and the LY6/uPAR family of cysteine-rich proteins characterized by a three-finger LU domain[3][7][6][1]. Like related LY6 proteins, it is primarily localized at the cell surface and is involved in immune cell differentiation and immune signaling, with evidence for roles in inflammatory processes[3][1][6]. Comparable LY6 family members, such as G6f proteins within the MHC region, are glycosylated cell-surface receptors that undergo tyrosine phosphorylation and interact with adaptor proteins Grb2 and Grb7, linking LY6G6F to intracellular signaling pathways involved in immune activation[3][6]. Expression of LY6G6F is upregulated in leukocytes in specific pathological states, including frontotemporal lobar degeneration associated with GRN mutations, suggesting a possible role in neuroinflammation or neurodegenerative disease progression[3]. The protein's structure, rich in disulfide bridges, forms a three-finger motif adaptable for various protein-protein interactions[5][1].
