Overview

Lymphocyte antigen 6 family member D (LY6D), historically known as the E48 antigen, is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein predominantly expressed in squamous epithelia and associated squamous cell carcinomas. It belongs to the Ly-6/uPAR family and is localized to human chromosome 8q24.3. LY6D is involved in keratinocyte cell-cell adhesion and marks the earliest stage of B-cell specification, distinguishing between B and T cell lineages during lymphocyte development. It has high clinical relevance as a therapeutic and diagnostic target for antibody-based therapy in squamous cell carcinoma, especially of the head and neck, due to its restricted expression profile and functional involvement in tumor biology[1][2][3][4][5][7][9][10].
