Overview

Lymphocyte antigen 6 family member G5B (LY6G5B) is a human protein encoded by the LY6G5B gene, which is located in the major histocompatibility complex (MHC) class III region on chromosome 6[1][2]. LY6G5B is part of the Ly6/uPAR superfamily of cysteine-rich proteins, which typically feature a three-fingered LU domain. Unlike many family members that are GPI-anchored cell-surface proteins, LY6G5B appears to be a secreted protein and may act as a ligand for receptors on undifferentiated hematopoietic cells, suggesting roles in cell–cell communication and regulating hematopoietic cell differentiation[2][4][6]. The LY6G5B locus is subject to complex post-transcriptional regulation, and genetic studies have associated the broader region with autoimmune disease susceptibility, although direct functional links remain uncertain[2]. LY6G5B has also been implicated in Poirier-Bienvenu neurodevelopmental syndrome[1]. There are no reports of LY6G5B as a direct therapeutic target nor of drugs interacting with this protein.
