Overview

Lymphocyte antigen 6 family member G6D is a cell surface protein encoded by the LY6G6D gene located within the major histocompatibility complex (MHC) class III region on chromosome 6. It is part of the Ly‑6/uPAR superfamily characterized by one or more LU domains that form a three-fingered protein structure stabilized by conserved disulfide bonds. The protein is typically attached to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor. LY6G6D is predominantly expressed in myeloid lineage cells, including monocytes, macrophages, granulocytes, and neutrophils—especially during their development and differentiation. Its expression persists in mature granulocytes and circulating neutrophils. Functionally, LY6G6D plays an important role in modulating immune responses through its influence on neutrophil infiltration and migration. It has also been implicated as a selectively expressed tumor antigen for human microsatellite-stable colorectal cancers—making it an emerging target for immunotherapeutic strategies such as T-cell engagers aimed at redirecting T-cell cytotoxicity toward cancer cells expressing this molecule[5][6]. There are currently no widely recognized drugs directly targeting this molecule nor established biomarkers or safety concerns specifically associated with its therapeutic targeting. The gene has several known aliases reflecting its discovery history across different research contexts but consistently refers to the same molecular entity involved primarily with immune system regulation at cellular interfaces.
