Overview

Lymphocyte antigen 6 family member G6F-G6D fusion protein (LY6G6F-LY6G6D) is a cell surface protein encoded by a naturally occurring readthrough transcript between the LY6G6F and LY6G6D genes on chromosome 6[5][7]. The fusion protein shares sequence with both parental molecules and is anchored in the membrane by a GPI linkage, characteristic of the Ly6/uPAR superfamily[1][3]. It participates in modulating immune responses, particularly in myeloid cells such as neutrophils, and is implicated in the development of immune resistance and metastasis in colorectal cancer[1][3][4]. Its expression is regulated by the JAK-STAT pathway and can be targeted by bispecific antibodies in preclinical cancer models, making it a promising therapeutic target for immune modulation and cancer treatment[4][5]. Biological roles also include biomarker potential for tracking immune aging and dysfunction, but it presents notable safety challenges due to its broad expression in immune cell compartments[1][3].

Bispecific T-cell dependent antibody: redirects T-cells to target LY6G6D-positive tumor cells, triggering immune-mediated cytotoxicity against colorectal cancer. JAK/STAT pathway inhibitors: reduce LY6G6D-associated immune resistance and tumor growth in CRC.
