Lymphocyte antigen 6 family member S (LY6S) is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein in the Ly6/uPAR superfamily, characterized by a single LU domain structure. This IFN-inducible protein is encoded by the LY6S gene and displays multiple isoforms, primarily associated with the plasma membrane or secreted forms. LY6S is involved in immune signaling and is most highly expressed in immune-related human tissues, such as the spleen and in IFN-activated environments. Although specific physiological and pathological roles of LY6S are still emerging, related Ly6 family members are markers of immune cell subpopulations and are implicated in immune cell maturation, innate and adaptive immune responses, cell adhesion, and possibly in the regulation of inflammation and tumorigenesis. Dysregulation or overexpression of Ly6 proteins, including possible analogs of LY6S, has been linked to tumor immune resistance, notably in colorectal cancer, suggesting therapeutic potential as surface targets for immunotherapy or as biomarkers for patient stratification and monitoring.
