Lymphocyte antigen 6 complex locus D (LY6D) is a member of the Ly6/uPAR superfamily of cell-surface proteins, typically identified as a 22-kDa GPI-anchored antigen. It is highly and selectively expressed in stratified squamous and transitional epithelia and serves as a differentiation marker at critical branch-points of B- and T-lymphocyte development as well as in distinguishing between epithelial and non-epithelial cells. LY6D (also called E48 antigen) is clinically relevant as a cell-surface marker for tumor imaging in head and neck squamous cell carcinoma and is under study for its broader roles in cancer and immune biology. Its restricted tissue expression and cell-surface accessibility make it an attractive target for imaging and potentially therapeutic antibody-based approaches.
Monoclonal anti-E48 antibody: Binds specifically to LY6D antigen on tumor cells, allowing for selective imaging or targeting of squamous cell carcinoma in the head and neck with technetium-99m-labeled E48 IgG or F(ab')2. This enables detection and potentially therapy by targeting cell-surface antigen-expressing malignant cells
