Lymphocyte cytosolic protein 2 (LCP2), also known as SLP-76, is an adaptor protein expressed primarily in hematopoietic cells. It serves as a critical scaffold organizing multi-protein complexes downstream of immune receptor engagement in T cells, natural killer cells, mast cells, and platelets. In T cells, LCP2 links phosphorylated receptors to downstream effectors, mediating signaling cascades that include calcium mobilization, MAP kinase activation, integrin clustering, and transcription factor activation, all essential to T cell activation and immune synapse formation. LCP2 also participates in platelet aggregation and vascular development, with genetic deficiency causing severe immunological and vascular defects in animal models. No small molecule or approved drug is currently known to selectively target LCP2 directly, but its expression and phosphorylation state are used as biomarkers of immune cell activation[1][2][3][4].
