Lck is a Src family non-receptor tyrosine kinase crucial for T-cell development and activation. It initiates and propagates T-cell receptor (TCR) signaling by phosphorylating ITAMs on CD3/TCR complex subunits, leading to downstream signaling cascades and cytokine production. Lck activity is tightly regulated by phosphorylation at key residues (Y394, Y505) and conformational changes. Dysregulation of Lck can contribute to immune dysfunctions and hematological malignancies, making it a therapeutic target.
