Lymphotoxin-alpha (LT-α)

Target
LT-α
Molecular classification
Cytokine, Tumor Necrosis Factor Superfamily Ligand
01

Overview

Lymphotoxin-alpha (LT-α), also known as tumor necrosis factor-beta (TNF-β), is a cytokine belonging to the tumor necrosis factor superfamily. It plays a critical role in immune regulation, inflammation, and lymphoid organ development. It exists as a soluble homotrimer or forms heterotrimers with lymphotoxin-beta (LTβ). LTα interacts with several receptors, including TNFRSF1A/TNFR1, TNFRSF1B/TNFBR, HVEM/TNFRSF14, and LTβR/TNFRSF3, triggering signaling pathways involved in inflammation, apoptosis, immune cell activation, and tissue organization.

Safety considerations

Overexpression or deficiency can influence cancer progression variably, sometimes promoting or inhibiting tumor growth/metastasis.Potential for off-target effects due to interactions with multiple receptors.Implication in autoimmune diseases; resistance to anti-TNF therapies may require targeting LTα instead.
06

Biomarkers

Genetic variations in LTA are linked to disease susceptibility and prognosis in myelodysplastic syndromes.
