Overview

LYPLAL1 antisense RNA 1 (LYPLAL1-AS1) is a long noncoding RNA (lncRNA) transcribed antisense to the LYPLAL1 gene on chromosome 1 and is 523 nucleotides in length. It is primarily cytoplasmic in human adipose-derived mesenchymal stem cells and is strongly upregulated during adipogenic differentiation. LYPLAL1-AS1 regulates this process by targeting the protein desmoplakin, modulating its stability (likely via the proteasome), resulting in inhibition of the Wnt/β-catenin signaling pathway, and promoting adipocyte lineage commitment. Its activity suggests an important role in adipogenesis and potential as a therapeutic target for obesity and related metabolic disorders, though there are currently no direct therapeutic agents targeting this RNA. The predominance in the cytoplasm and specific upregulation during adipogenesis underscore its functional significance in fat cell development

Mechanism of action

No direct drugs, but manipulation (e.g., knockdown or overexpression) of LYPLAL1-AS1 alters adipogenic differentiation through modulation of desmoplakin protein stability and inhibition of the Wnt/β-catenin pathway

03

Biological functions

Regulation of adipogenic differentiationModulation of mesenchymal stem cell lineage fatePost-transcriptional regulation of protein stability (notably desmoplakin)Inhibition of Wnt/β-catenin signaling pathway
04

Disease associations

ObesityAdipogenesis-related disordersPotentially other disorders related to abnormal fat cell development
05

Safety considerations

Targeting lncRNAs may present delivery and specificity challenges due to their nuclear/cytoplasmic localization and poor sequence conservation across speciesOff-target effects on vital differentiation pathways in stem cells should be considered
06

Biomarkers

LYPLAL1-AS1 expression may serve as a biomarker for adipogenic differentiation potential or abnormal adipogenesis
