Lysine-rich nucleolar protein 1 (KNOP1) is a nucleolar protein encoded by the KNOP1 gene located on chromosome 16p12.3. It is highly expressed in the testis, superior cervical ganglion, placenta, and developing heart and lung, and shows lower expression in adult liver and pancreas[1][2][6]. The protein contains lysine-rich regions and two main domains: DUF5595 (sometimes absent in specific isoforms) and SMAP. KNOP1 is believed to interact with ZNF106 and is hypothesized to play a role in processes similar to those of nucleostemin, such as cell proliferation and development. It has been proposed as a prognostic biomarker in hepatocellular carcinoma, where high expression correlates with worse outcomes[6]. Although its exact molecular function remains unclear, elevated KNOP1 levels are associated with cell cycle regulation and cancer progression. No drugs are known to target KNOP1 directly, and there are no established mechanisms of action or safety concerns documented for therapeutics involving this protein[1][2][6].
