Overview

LysM domain-containing proteins are a family of proteins characterized by the presence of one or more Lysin Motifs (LysM), which serve as highly conserved carbohydrate-binding modules. These domains primarily recognize and bind to 1,4-beta-linked N-acetylglucosamine polymers, such as peptidoglycan in bacterial cell walls and chitin in fungal cell walls (UniProt, 2023). In humans, members of this family, including LYSMD1, LYSMD2, LYSMD3, and LYSMD4, play significant roles in the innate immune system by acting as pattern recognition receptors that detect microbial components and trigger inflammatory signaling (PubMed: 25130630). Beyond immunity, certain members like LYSMD4 have been implicated in cancer progression, specifically in colorectal cancer, where they may influence cell proliferation and migration (PubMed: 31233145). While there are currently no FDA-approved drugs specifically targeting human LysM domain-containing proteins, they are considered emerging targets for treating infectious diseases and modulating immune responses in chronic inflammation. Therapeutic strategies under investigation include the development of small molecule inhibitors or monoclonal antibodies designed to block the carbohydrate-binding site, thereby preventing the activation of pro-inflammatory cascades.
