Overview

LysM RLK1-interacting kinase 1 (LIK1) is a leucine-rich repeat receptor-like kinase (LRR-RLK) primarily characterized in Arabidopsis thaliana. It serves as a critical component of the plant's innate immune system by regulating microbe-associated molecular pattern (MAMP)-triggered immunity. LIK1 physically interacts with the chitin receptor CERK1 and the extracellular ATP receptor P2K1, acting as a molecular hub that integrates various stress signals [1, 4]. While it negatively regulates chitin and flagellin signaling to prevent over-activation of the immune response, it positively modulates the response to extracellular ATP, which acts as a damage-associated molecular pattern (DAMP) [3, 4]. Mutations in LIK1 lead to enhanced resistance against hemibiotrophic pathogens like Pseudomonas syringae but increased susceptibility to necrotrophic fungi such as Sclerotinia sclerotiorum, highlighting its complex role in balancing different defense pathways [1, 5].

Mechanism of action

LIK1 acts as a negative regulator of chitin-induced and flagellin-induced immune responses by interacting with and being phosphorylated by the chitin receptor CERK1 [1, 3]. It also functions as a positive regulator of extracellular ATP (eATP) signaling through its physical interaction with the eATP receptor P2K1 [4].
