Overview

Lysosomal-associated membrane protein 5 (LAMP5), also known as BAD-LAMP, is a type I transmembrane glycoprotein primarily localized to the lysosomal membrane [1, 2]. While its physiological expression is largely restricted to the central nervous system and plasmacytoid dendritic cells, it has been identified as a critical survival factor in specific hematologic malignancies, including blastic plasmacytoid dendritic cell neoplasm (BPDCN) and MLL-rearranged leukemias [3, 4]. In these contexts, LAMP5 functions to modulate autophagy and proteasomal degradation pathways, preventing the turnover of oncogenic proteins [4, 5]. Therapeutic intervention strategies are currently focused on the use of antisense oligonucleotides (ASOs) to target and degrade LAMP5 mRNA, which has demonstrated the ability to inhibit tumor growth in preclinical models [5]. Given its restricted expression profile, LAMP5 is considered a high-priority target for precision oncology, although its role in the brain necessitates careful evaluation of potential neurotoxicities [1, 5].

Mechanism of action

Antisense oligonucleotide-mediated degradation of LAMP5 mRNA via RNase H-dependent cleavage, leading to reduced protein expression and subsequent induction of apoptosis in malignant cells [4, 5].
