Overview

Lysosomal membrane permeabilization (LMP) is a cellular event in which the integrity of the lysosomal membrane is compromised, allowing the release of lysosomal contents—primarily hydrolytic enzymes such as cathepsins—into the cytosol. This process can trigger various forms of cell death, including apoptosis and necrosis, and plays a significant role in both physiological regulation and disease pathology. LMP can be induced by reactive oxygen species (ROS), lysosomotropic agents, endogenous proteins like Bax, and cellular stressors. Regulatory factors include lipid composition of lysosomal membranes, p53, Bcl-2 family proteins, and the TORC1 signaling pathway. Dysregulation or increased susceptibility to LMP has been implicated in cancer cells' resistance/sensitivity to therapy.
