Lysosome-associated membrane glycoprotein 3 (LAMP3) is a heavily glycosylated type I transmembrane protein mainly localized on lysosomal and lysosome-related organelle membranes. Primarily expressed in mature dendritic cells in humans, LAMP3 is associated with the regulation of antigen processing and presentation in the immune response, particularly within the MHC class II compartment[1]. In the lung, LAMP3 is also highly expressed in alveolar type II cells where it localizes to lamellar bodies, important for pulmonary surfactant metabolism and homeostasis[2]. The protein has been linked to the maturation of dendritic cells, resistance to certain viral infections, modulation of apoptosis (under specific conditions, e.g., salivary gland or tumor cells), and the proliferation and survival of pathogens like Salmonella within host cells[3][4]. LAMP3 is considered a marker for dendritic cell maturation and is involved in autoimmune and respiratory diseases as well as various cancers, where it may promote metastasis or drug resistance. Although there are no therapeutics directly targeting LAMP3 as of the current knowledge, its disease associations and cell biology roles make it a candidate of interest for future drug development[1][3][4].
