Overview

Lysosome-associated membrane glycoprotein 5 (LAMP5) is a member of the LAMP family that is distinct from canonical members like LAMP1 and LAMP2 in its expression and localization. Unlike other LAMPs, LAMP5 is predominantly and specifically expressed in subpopulations of GABAergic neurons in the brain, where it localizes at synaptic terminals and is implicated in the fine-tuning of short-term synaptic plasticity and the regulation of evoked GABAergic neurotransmission. Alterations in LAMP5 function influence synaptic transmission dynamics and are associated with subtle behavioral phenotypes, including decreased anxiety and olfactory discrimination deficits in knockout mice. In *C. elegans*, the ortholog UNC-46 is a trafficking chaperone for the vesicular GABA transporter; in mammals, LAMP5 appears to play a synaptic regulatory rather than general trafficking role. Human disease associations include epilepsy and neurodegenerative disorders such as familial temporal lobe epilepsy and frontotemporal dementia. There are currently no known drugs that target LAMP5, and it is not considered a therapeutic target at this time[1][3][6][7].
