Overview

Lysyl oxidase (LOX) is a copper-dependent amine oxidase enzyme that plays a central role in the biogenesis and maintenance of connective tissue matrices. Its primary function is to catalyze the oxidative deamination of specific lysine residues in collagen and elastin precursors, converting them into reactive aldehydes (allysine). These aldehydes then participate in covalent cross-linking reactions, which are essential for stabilizing the extracellular matrix (ECM) by forming mature collagen fibrils and elastic fibers. LOX expression is regulated by various factors, and dysregulation has been linked with tumor progression due to effects on cell migration/invasion via ECM remodeling. Inhibition of LOX is explored as a strategy to treat fibrotic diseases and cancer.
