Overview

LZTS1 antisense RNA 1 (LZTS1-AS1) is a long non-coding RNA (lncRNA) transcribed antisense to the LZTS1 gene. It does not code for a protein and is predominantly located in the nucleus of colorectal cancer (CRC) cells[1][3]. LZTS1-AS1 is frequently upregulated in CRC, where its increased expression correlates with a more advanced tumor stage, metastasis, and worse prognosis. Mechanistic studies show that LZTS1-AS1 interacts with and destabilizes LDLRAD4 mRNA, resulting in reduced LDLRAD4 expression. This molecular effect promotes epithelial-to-mesenchymal transition (EMT) and enhances the migratory and invasive properties of CRC cells, thereby facilitating metastasis[1]. Its expression profile and functional impact suggest that LZTS1-AS1 may act as a prognostic biomarker and a potential therapeutic target in colorectal cancer, though no direct pharmacological agents against it are in clinical use[1][3]. As a representative of antisense lncRNAs, LZTS1-AS1 exemplifies complex gene regulatory mechanisms affecting tumor biology[2].

Mechanism of action

No direct pharmacological target for drugs has been established to date. For experimental targeting, silencing LZTS1-AS1 leads to upregulation of LDLRAD4, reducing EMT and metastasis in colorectal cancer.

Biological functions

Disease associations

CancerColorectal cancer (CRC): associated with disease progression, metastasis, and prognosis
05

Safety considerations

No specific safety concerns reported, as LZTS1-AS1 is not currently a drug target in therapy[1][3].General concerns for targeting lncRNAs may include specificity and off-target gene expression effects[2].
06

Biomarkers

High expression level of LZTS1-AS1 in tumor tissue is a biomarker for poor prognosis and advanced stage colorectal cancer[1].Potential biomarker for selection and monitoring in CRC clinical research[1].
