M-phase phosphoprotein 6 (MPHOSPH6 or MPP6) is a nucleolar protein that binds RNA, especially pyrimidine-rich sequences in the internal transcribed spacer 2 (ITS2) region of pre-ribosomal RNA. Its principal role is to recruit the exosome complex for rRNA processing, specifically the generation of the mature 3′ end of 5.8S rRNA, in the context of ribosome biogenesis. It lacks intrinsic exonuclease or helicase activity, instead functioning as a cofactor and adapter for the exosome. MPHOSPH6 accumulates in nucleoli, associates with high-molecular-weight pre-ribosomal complexes, and is critical for precise rRNA maturation. Studies show that MPP6 knockdown impairs the processing of 5.8S rRNA, demonstrating its importance for ribosome assembly in human cells.
