Mab-21 domain-containing protein 2 (MB21D2) is a member of the cGAS/DncV-like nucleotidyltransferase (CD-NTase) family and acts as a modulator of the cGAS-STING innate immune signaling pathway[1]. It physically interacts with and enhances cGAS activation, promoting liquid phase condensation, DNA binding, and cGAMP synthesis in response to DNA stimulation, leading to increased type I interferon production and antiviral/antitumor immunity[1]. MB21D2 is also implicated in cell fate determination, organogenesis, and cancer biology, particularly as a signaling hub connecting cadherin-binding, cell proliferation, migration, and epithelial-mesenchymal transition pathways; recurrent mutations (e.g., Q311E) and overexpression are found in head and neck squamous cell carcinoma, suggesting a role in cancer progression and stemness[2].
Enhancement of cGAS enzymatic activity, leading to increased production of cyclic GMP-AMP (cGAMP), which activates interferon signaling. Possible scaffolding or adaptor function linking cadherin/E-cadherin and other signaling pathways.
