Overview

MACC1 antisense RNA 1 (MACC1-AS1) is a long non-coding RNA (lncRNA) transcribed from the sixth intron of the MACC1 gene, known for its role in regulating gene expression rather than coding for proteins. MACC1-AS1 regulates the transcription of the oncogene MACC1 via two distinct mechanisms. In the nucleus, it acts as a cis-acting lncRNA by forming a complex with the RNA-binding protein DDX5, which interacts with the distal region of the MACC1 promoter, facilitating DDX5 and SP-1 recruitment to the promoter and thus enhancing MACC1 expression. In the cytoplasm, MACC1-AS1 functions as a competing endogenous RNA (ceRNA), “sponging” tumor suppressor miRNAs (e.g., miR-384, miR-145-3p), indirectly regulating the levels of target mRNAs like c-Myc and pleiotrophin. MACC1-AS1 is upregulated in multiple cancer types, especially breast and gastric cancers, and its expression is associated with increased tumor proliferation, invasion, and poor clinical outcome. No approved drugs currently target MACC1-AS1, and it is not considered a classical therapeutic target like a protein receptor or enzyme, but rather a potential biomarker or experimental target for anti-cancer strategies[1][2].
