Overview

Macoilin (MACO1) is a highly conserved multi-transmembrane protein expressed specifically in the nervous system, mainly localized to the rough endoplasmic reticulum (rER) in neurons[1][2]. Macoilin is required for various neuronal functions, including normal locomotion, chemotaxis, and thermotaxis in *Caenorhabditis elegans*, and plays a role in the regulation of neuronal excitability[1][2]. The protein is believed to facilitate protein folding, assembly, and traffic of membrane proteins, potentially including ion channels essential for neural activity[2]. While its broad conservation from invertebrates to mammals suggests an important neural role, there is currently no evidence supporting its direct involvement as a therapeutic target, involvement in human disease, or interaction with drugs[1][2]. **Special note:** - TMEM57 is sometimes used as a synonym, but this is potentially incorrect; TMEM57 in humans is generally annotated as a distinct protein unrelated to nervous system macoilins. Most functional data regarding MACO1 comes from non-human models (*C. elegans*, mouse)[1][2]. - Macoilin is not classified as a receptor, enzyme, transporter, or standard drug target category, and there is no established therapeutic targeting or drug modulation known[1][2].
