Overview

MACROD2 antisense RNA 1 (MACROD2-AS1) is a long non-coding RNA (lncRNA) transcribed from the antisense strand of the MACROD2 gene locus on chromosome 20. Its primary function is believed to be the regulation of MACROD2 gene expression, rather than coding for a protein[2]. Expression profiling across tissues shows that MACROD2-AS1 is present in various adult and fetal tissues, sometimes at higher levels than its sense counterpart MACROD2[4]. A genetic association signal for autism spectrum disorder (ASD) maps near the MACROD2 locus, and MACROD2-AS1 expression is significantly elevated in the temporal cortex of individuals with ASD, suggesting a possible role in neural development or disease risk[4]. However, mechanistic or therapeutic targeting information is lacking, and no direct interaction with drugs has been reported. MACROD2-AS1 should not be considered a druggable target such as a receptor, enzyme, or transporter; it is classified as a regulatory RNA with yet-to-be-elucidated biological significance[2][4].
