Macromolecular synthesis refers to the fundamental biological processes by which cells produce large, complex molecules such as DNA, RNA, and proteins. These processes—replication, transcription, and translation—are essential for cell survival, growth, and reproduction, as they facilitate the flow of genetic information and the production of structural and functional cellular components (Nature Education, 2014). In the context of pharmacology, macromolecular synthesis is not a single molecular target but rather a broad category of pathways targeted by various therapeutic agents. For example, antimetabolites like methotrexate and 5-fluorouracil interfere with DNA synthesis to treat cancer, while various antibiotics like tetracycline and rifampicin target bacterial macromolecular machinery to treat infections (StatPearls, 2023; NCBI Bookshelf, 2021). Because these processes are conserved and vital for normal cell function, drugs targeting macromolecular synthesis often exhibit significant side effects, such as myelosuppression and mucosal damage, due to their impact on rapidly dividing healthy tissues (PubMed, 2022).
