Macromolecular synthesis and metabolic pathways encompass the complex network of chemical reactions that allow cells to produce essential biological polymers—DNA, RNA, and proteins—and generate energy (NIH, 2023). These processes are fundamental to cellular homeostasis, growth, and division, involving both catabolic pathways that break down nutrients and anabolic pathways that build cellular components (StatPearls, 2023). Because these pathways are also vital for healthy cells, drugs targeting them frequently cause systemic toxicities, such as bone marrow suppression and mucosal damage (NIH, 2023). This entry is considered incorrect as a single therapeutic target because it describes a broad set of biological processes rather than a specific molecular entity.
Drugs targeting these pathways typically act as antimetabolites that mimic natural substrates to inhibit rate-limiting enzymes or as inhibitors of the machinery responsible for polymerizing nucleic acids and proteins.
