Overview

The term macrophage-associated protein refers to a broad class of proteins expressed by or associated with macrophages, which are key innate immune cells involved in phagocytosis, antigen presentation, and cytokine production. These proteins often serve as markers for macrophage polarization, such as CD163 and CD206 for M2-like (anti-inflammatory/pro-tumor) macrophages and HLA-DR or iNOS for M1-like (pro-inflammatory/anti-tumor) macrophages. In therapeutic contexts, macrophage-associated proteins are targeted to modulate the immune microenvironment, particularly in cancer, where tumor-associated macrophages (TAMs) often promote tumor growth and immunosuppression. Drugs targeting these proteins, such as antibodies against SIGLEC-15, TREM2, or the CD47-SIRPα axis, aim to reprogram macrophages to enhance anti-tumor immunity or inhibit their immunosuppressive functions. Due to the diversity of proteins in this category, the specific biological role and therapeutic potential depend on the individual molecule being targeted.
